Reviewed by ICLAC (ref: 191110). MGC-803 was reported to originally come from gastric carcinoma but several papers from cell repositories in China have shown that it has a high similarity to HeLa. Looking at the STR profiles that we have available, they appear to fall into two groups. One is consistent with HeLa, while the other has additional alleles at many loci e.g. Penta E has "7,10,16,17". This second group of samples may represent a mixture of HeLa and a second cell line; it may also represent a hybrid cell line (e.g., following exposure to a fusogenic agent).

Instruction

Use exclusive Red Cotton system to design a sgRNA fragment knockout strategy for the DSC2 gene, clone the two sgRNAs into CRISPR-U™ vector, transfect the vector into the MGC80-3 cell line by electroporation/lentivirus method, use the limited dilution method for single-cell cloning and use the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000) for nucleic acid lysis and PCR amplification on the single-cell clones, followed by Sanger sequencing to verify the genotype. Upon passing the verification, DSC2 gene knockout DSC2 Knockout cell line (MGC80-3) will be expanded and cryopreserved.
