1. Technical Details:
Ubigene uses Tol2 transposon system to construct transgenic Zebrafish. The transposon system includes transposon and transposase. The target expression frame is located between the two transposons of the expression plasmid. The transposase plasmid is transcribed into mRNA in vitro and co-injected with the expression plasmid. The transposase can cut the sequence between the two transposons and paste it into the genome of zebrafish zygote. After genotyping, the individuals with successful integration of target gene will be selected for passage.
Construction of Transgenic Zebrafish Using Tol2 Transposition System
2. Key Features:
Ubigene uses Tol2 transposon system, which has high integration efficiency and short turnaround.
3. Applications:
Drug screening; Disease models; Gene function study.
※In addition to Zebrafish gene editing, Ubigene also provides services of compound screening and evaluation of efficacy and safety.
Screening of small molecule compounds has always been the focus of research. Traditional screening methods include in vitro screening (bacteria, yeast, mammalian cells) and invertebrate model screening (drosophila, nematode). In vitro screening cannot simulate the micro-environment in vivo, and some special types of cells and most organs can not be cultured or reconstructed in vitro. The invertebrate model screening lacks complex systems and organs, and its morphology and structure are far from that of human beings. So these two screening methods have limitations. Zebrafish, as a model animal for compound screening, provides a complete living vertebrate system with strong reproductive capacity and low feeding cost, which combines the biological complexity of in vivo models with the ability of high-throughput screening.
Ubigene have a lot of experience on drug efficacy and safety assessment of zebrafish. Common screenings are as follows:
· Drug Screening for Cardiovascular Diseases
· Screening of Antineoplastic Drugs
· Drug Screening for Neurological Diseases
· Drug Screening for Liver Diseases
· Study of Whitening Function of Cosmetics
· Evaluation of drug trans-blood-brain barrier
· Acute Toxicity Assessment of Compounds
· Chronic Toxicity Assessment of Compounds
· Cardiovascular Toxicity Assessment of Compounds
· Hepatotoxicity Assessment of Compounds
